乳酸(LA)的过量产生可能发生在运动期间或者许多疾病中,例如癌症中。个人伴有高乳酸血症的患者常表现为贫血、血清铁减少以及一种铁代谢关键调控因子—铁调素(hepcidin)升高。然而,目前尚不清楚乳酸是否以及如何调节铁调素的表达。最近,中国科学院生态环境科学研究中心在期刊《Cell Metabolism》(IF=29)上发表题为“Lactate modulates iron metabolism by binding soluble adenylyl cyclase.”的文章,展示乳酸与正常肝细胞中的可溶性腺苷酸环化酶(sAC)结合,并影响正常肝细胞中的全身铁稳态 上调小鼠铁调素的表达。全面的体外、体内和计算机实验表明 LA-sAC相互作用提高了环磷酸腺苷(cAMP)水平,从而激活PKA-Smad1/5/8信号通路,增加铁调素转录。同时在野生型小鼠和铁稳态紊乱的小鼠中验证了这一调节轴。LA也调节人体静息状态下和经受大量运动条件下的铁调素。该研究将高乳酸血症与缺铁联系起来,为运动员和乳酸酸中毒患者的贫血提供了一个全新的机制的阐述。
