疫苗诱导的抗肿瘤免疫应答速率过慢无法有效抑制恶性肿瘤的快速生长,常常限制了肿瘤治疗性疫苗的效率,如何快速诱导并维持强烈的抗肿瘤免疫应答是当前肿瘤治疗性疫苗研究待解决的难点之一。许多功能化的微/纳米疫苗被证明能加强肿瘤免疫治疗,但很少有研究将这些微/纳米疫苗的治疗效果与疫苗接种途径(如静脉注射、皮下注射或两者的组合)联系起来。近日,中国科学院过程工程研究所刘瑞田研究员、昆士兰大学澳大利亚生物工程与纳米技术研究所许志平教授和张凌宵博士等人在《Nano Today》(IF=16.907)上发表了题为”Nanovaccine’s rapid induction of anti-tumor immunity significantly improves malignant cancer immunotherapy”的研究论文,首次提出联合静脉免疫(IV-VAC)和皮下免疫(SC-VAC)的肿瘤疫苗接种新策略,成功实现强烈的抗肿瘤免疫应答的快速诱导与维持。本文采用经典的启动+促进免疫策略,研究了4种纳米疫苗组合(IV+IVIV+SCSC+IVSC+SC)对E.G7-OVA淋巴瘤和B16F10黑色素瘤小鼠抗肿瘤治疗效果的影响。通过将抗原和CpG加载到层状双金属氢氧化物(Layered Double Hydroxide, LDH)纳米粒子上构建纳米疫苗。实验数据表明,采用“IV初免 + SC加强IV + SC)的疫苗接种策略(使用IV-VAC进行初次免疫,随后使用SC-VAC进行免疫加强),能够在肿瘤早期快速诱导机体产生强烈的抗肿瘤免疫应答来抑制肿瘤生长,并在SC-VAC的帮助下逐步加强免疫强度。
