Cayman’s sPLA2 (Type V) Inhibitor Screening Assay Kit provides a robust and easy-to-use platform for identifying novel inhibitors of human sPLA2 type V. The assay uses diheptanoyl thio-PG, a sulfur-containing analog of phosphatidylglycerol, which is the preferred substrate of sPLA2 type V. Upon hydrolysis by sPLA2 type V at the sn-2 position, the free thiol can be detected by the sPLA2 Assay Probe. This results in a product that can be easily quantified by measuring absorbance at a wavelength of 410 nm. A potent and reversible inhibitor, LY315920 (included in this kit as sPLA2 Control Inhibitor), inhibits the enzyme with an IC50 value of approximately 100 nM.
