The components of this kit can be combined to produce the small molecule cocktail CEPT. When mixed with the appropriate cell culture media, CEPT promotes cell survival and cryopreservation of genetically stable human pluripotent stem cells (hPSCs).{55519,71300} Chroman 1, a Rho-associated kinase (ROCK) pathway inhibitor, blocks cell contraction, which is detrimental to dissociated hPSCs. Emricasan synergistically improves cell survival through its action as a pan-caspase inhibitor, and trans-ISRIB regulates protein translation by directly targeting eukaryotic initiation factor 2β (eIF2β) to ensure unperturbed protein synthesis. Polyamines Supplement Mix contains essential polyamine cations known to support cell growth.Each component in this kit is provided separately, and instructions are included to reconstitute and mix these reagents for direct dilution into cell culture media. The kit is intended to supplement up to 3 L of culture media when following the supplied instructions. Some reagents may be provided in excess and may be properly disposed of after cell media preparation. Prior to use, reagents must be stored as instructed at -20°C.
