2025年12月12日,香港中文大学王东安教授团队在《ADVANCED MATERIALS》(IF 26.8)上发表题为 “ 3D Bioprinted Human Synovium-Cartilage Models Mimic Rheumatoid Arthritis Microenvironment and Recapitulate In Vivo Therapeutic Responses ” 的研究论文,构建了一种高度仿生的3D生物打印人滑膜-软骨模型。该模型可有效模拟类风湿关节炎(RA)的微环境,并重现体内治疗反应,为RA致病机制研究和临床前药物筛选提供了有力工具。

ADVANCED MATERIALS


类风湿关节炎(RA)模型在治疗发现和基础研究中发挥着关键作用。然而,现有模型在准确模拟体内微环境方面存在局限性,并且缺乏复杂的细胞间相互作用。在此,本研究提出了一种体外RA模型,该模型能够真实复刻体内软骨和滑膜衬里的功能和组成特性,通过将软骨细胞再细胞化到II型胶原蛋白支架上,并利用3D生物打印技术构建包含成纤维样滑膜细胞(FLS)和促炎性巨噬细胞的顶层以及保护屏障巨噬细胞的底层的双层明胶-基质胶水凝胶来建立。


这种滑膜—软骨系统重现了类似于RA的关键炎症过程,包括促炎介质和降解酶的产生增加、活性氧的生成、FLS向软骨的侵袭、巨噬细胞表型的改变以及软骨细胞外基质成分的耗竭。所建立的模型能够有效地筛选抗关节炎药物,这一点通过使用celecoxib和tofacitinib得到了验证。此外,转录组和蛋白质组学分析证实,该模型能精准模拟体内病理状态。值得注意的是,与类风湿关节炎(RA)的大鼠模型相比,这种体外模型能够反映疾病对药物的应答反应。综上,这项研究为RA治疗药物的临床前筛选提供了可靠的体外人滑膜-软骨模型。
