Tyrphostin A9 is a well‑characterized inhibitor of PDGF receptor tyrosine kinase, exhibiting an IC₅₀ of 1.2µM. It also functions as a potent inhibitor of oxidative phosphorylation at nanomolar concentrations (10nM). Beyond its primary kinase‑related effects, Tyrphostin A9 suppresses cellular proliferation by disrupting mitochondrial integrity and perturbing Golgi complex organization. These actions collectively contribute to its ability to block the growth of vascular smooth muscle cells.
Of particular relevance to neuroscience, Tyrphostin A9 has been shown to induce mild mitochondrial uncoupling in midbrain dopaminergic (mDA) neurons. This mitochondrial modulation is associated with a reduction in intracellular α‑synuclein levels, suggesting a potential therapeutic angle for disorders characterized by α‑synuclein accumulation.
