Ergothioneine (EGT) is a naturally occurring amino acid derivative with potent antioxidant and cytoprotective properties. As a stable antioxidant, it scavenges reactive oxygen and nitrogen species, protects mitochondria, stabilizes DNA and proteins under oxidative stress and supports cellular defense mechanisms. Widely studied in the context of aging, neuroprotection, cardiovascular health, immune modulation, and skin protection, EGT is used in dietary supplements, functional foods, cosmetics and as a research reagent for studying redox biology and stress-related cellular pathways. Ergothioneine is transported by the organic cation/carnitine transporter 1, which has been linked with autoimmune diseases, including rheumatoid arthritis and Crohn’s disease. EGT, with its strong antioxidant effect, is widely used as an active ingredient for anti-aging cosmetics. Ergothioneine is also called a longevity vitamin. It accumulates in high levels in human tissues. In human plasma, EGT declines with age and reduced EGT levels are linked to a multitude of age-related disorders, including neurodegenerative and cardiovascular diseases, while EGT supplementation is protective in a broad range of disease and aging models. The mechanism of action has been recently deciphered. Upon exercice training, EGT accumulates in muscle mitochondria where it binds and activates 3-mercaptopyruvate sulfurtransferase (MPST), which boosts mitochondrial respiration and exercise training performance in mice. Increasing EGT in the diet increases exercise performance of mice by 19%, making EGT an important factor in health and aging.
